The present investigation an attempt has been made to increase therapeutic efficacy, reduce frequency of administration and improve patient compliance, by developing sustained release matrix tablets of Allopurinol. Sustained release matrix tablets of Allopurinol, were developed by using different drug: polymer ratio. Kollidon SR, Hydroxypropyl methylcellulose K15M, K100M as matrix former. All lubricated formulations were compressed by direct compression and by wet granulation method. Compressed tablets were evaluated for uniformity of weight, content of active ingredient, friability, hardness, thickness, in vitro dissolution, and swelling index. All the formulation showed compliance with pharmacopoeial standards. Among the different formulation, B8 showed sustained release of drug for 12 hours with 86.55% release. The regression coefficient value of Higuchi plot was found to be 0.9925 that showed that drug was released by diffusion mechanism. The slope value of korsmeyer ‐peppas equation was found to be 0.5062 which indicating that drug was released by non‐fickian release mechanism. The R2 value for Hixson Crowell plot was found to be 0.9919 which indicates that drug release was limited by drug particle dissolution rate and erosion of the polymer matrix. Thus, drug in combination with Hydroxy propyl methyl cellulose K100M were found to be effective in retarding the release of Allopurinol.Keywords

Asian Journal of Pharmaceutical Analysis, Vol 6, No 3 (2016), Pagination: 167-182Abstract
The aim for the present study was to develop a delivery system wherein the retention of ofloxacin could be achieved for increasing local action in gastric region against Helicobacter pylori. Therefore the present investigation is concerned with the development of rice bran oil entrapped zinc pectinate beads containing ofloxacin, which after oral administration were designed to prolong the gastric residence time, thus to increase the bioavailability of the drug. A suitable method of analysis of drug by UV spectrophotometry was developed. Ofloxacin showed maximum absorption at a wavelength 294.5 nm in pH 1.2 hydrochloric acid buffer. The value of regression coefficient (r2) was found to be 0.999, which showed linear relationship between concentration and absorbance. Preformulation study for drug and polymer compatibility by DSC and FTIR gave confirmation about the purity of the drug and showed no interaction between drug and the polymers.Keywords

Bosentan is an endothelial receptor antagonist (ERA) indicated for the treatment of Pulmonary arterial hypertension (PAH). The aim of the present study involves the development of controlled release tablets of bosentan. The tablets were prepared to release the drug for a prolong period of time within the GIT, to enhance the bioavailability, to minimize the dosing frequency, and to improve the patient compliance. The tablets were formulated by using various polymers like hydroxyl propyl methyl cellulose, acacia and xanthum. The formulated tablets were evaluated. The drug release from the optimized formulation (F7) was found to be the best after observing the results of dissolution rate and pre & post formulation studies.Keywords

Asian Journal of Pharmaceutical Analysis, Vol 7, No 3 (2017), Pagination: 176-184Abstract
The objective of the study was to prepare and evaluate ACS contain pellets by the process of extrusion followed by spheronization for controlled release. The method employed was economical and does not imply the use of toxic solvents. The aqueous SLS which forms micropores on the surface of the pellets. The results of micromeritic properties, Hausner ratio and friability of the pellets were well within the limits which indicate good flow potential for the prepared pellets. Drug contains pellets exhibited spherical nature as evidenced by SEM photomicrographs and sphericity studies. From the FTIR and DSC studies, it was observed that there was no chemical interaction between the drug and polymers used indicates that drug was in stable form. The drug content study revealed uniform distribution of the drug in the pellets. The drug release rate was found vary among the formulations depending on the compositions of polymers used. The obtained dissolution data indicated that the drug release through the microporous polymeric membrane follows Fickian diffusion. Optimized formulation F5 and marketed product Aceton SR100 mg tablet showed similarity in drug release profile. Formulation F5 is an ideal formulation for once daily administration. From the present study, it can be concluded that the prepared matrix pellets demonstrate the potential use of SA/GPS/MCC blend for the development of controlled drug delivery systems for many water insoluble drug.Keywords
